A broad-spectrum immunedefense against infection.

Alopexx is a clinical-stage biotechnology company developing a vaccine and an antibody against PNAG, a surface molecule shared by more than 75 pathogens. Our aim is to reduce the world's reliance on antibiotics.

Antibiotics attack microbes directly, and microbes learn to resist them. Our therapeutics work through the body's own immune system instead.

Immune therapeutics are not associated with the development of resistance and are not excreted into the environment. That makes them a durable complement to the antibiotics hospitals depend on today.

The science

One target, shared by bacteria, fungi and parasites.

PNAG, or poly N-acetyl glucosamine, helps microbes stick to tissue and implanted devices and shields them from the immune system. Most vaccines target a coat unique to one strain. PNAG appears across an expanding list of pathogens.

Natural PNAG is heavily acetylated, which provokes a weak immune response. Our therapeutics target its de-acetylated backbone instead, inducing antibodies that kill PNAG-bearing microbes. The work began with Dr. Gerald Pier and his team at Harvard Medical School and Brigham and Women's Hospital.

Pipeline

Two clinical candidates. Four programs from one platform.

An antibody for immediate protection and a vaccine for lasting protection, plus related formulations for neuroinflammatory disease.

Fully human monoclonal antibody

F598

Designed to prevent and mitigate hospital-associated infections in ICU patients. A single infusion has provided antibody coverage for two to three months.

Phase 1 and pilot phase 2 completed. 1,600-patient phase 2 ICU study with AP-HP anticipated in Q1 2027.

Synthetic conjugate vaccine

AV0328

A complement to approved pneumococcal vaccines, which cover at most 23 of the 100 known serotypes.

Phase 1 completed in 16 healthy volunteers. Phase 2 dose optimization planned.
Neuroinflammation formulations

AB0411 and AV0407

Related antibody and vaccine formulations for Alzheimer's disease and other neuroinflammatory conditions.

Development anticipated to begin in Q1 2027.
Antimicrobial resistance
4.95M

deaths worldwide were associated with bacterial antimicrobial resistance in 2019, making it the third leading cause of death.

The Lancet, 2022 global burden analysis of 204 countries
"With the increasing incidence of antimicrobial resistant bacteria there is a pressing need for novel and alternative therapeutics."

Collaborations

Working with some of medicine's largest institutions.

ICU study partner

Assistance Publique, Hôpitaux de Paris

Europe's largest hospital system has agreed to run the 1,600-patient phase 2 study of F598 across eight of its hospitals.

Vaccine collaboration

Bharat Biotech

Co-development and commercialization of AV0328 in India and other low and lower-middle income countries.

Scientific origin

Brigham and Women's Hospital

Alopexx exclusively licenses issued patents covering the vaccine, the F598 antibody and their methods of use.

Neuroinflammation

Beyond infection, a new look at Alzheimer's disease.

PNAG has been found in the brains of patients with Alzheimer's disease and traumatic brain injury, where no live pathogen can be grown. We believe microbial fragments carrying PNAG drive chronic inflammation in these conditions.

In mouse models, vaccination against PNAG prevented cognitive decline. Our AB0411 and AV0407 formulations are designed to bring that approach to patients.
